preparation | The amino (4-methoxyphenyl) acetic acid was prepared as follows: the ketone (0.5mmol) and HCOONH 4(5mmol) was dissolved in MeOH(2ml) in a rotating disk reaction tube. The mixture was then degassed and stirred at 80 °c for 10 min under nitrogen. HCOOH / NEt3 azeotrope (0.5ml) and catalyst solution (1ml) were then introduced (prepared by dissolving catalyst 0.5 μmol in meh1oml). The resulting mixture was stirred at 80 °c for the indicated time. The reaction was quenched with water, basified with aqueous KOH and extracted with DCM. The solvent was then removed under vacuum. The crude product was dissolved in ethanol (10ml) and then 6N HCl solution (5ml) was added. The mixture was refluxed for 6 hours. The ethanol was then removed under vacuum and the resulting aqueous layer was washed with ethyl acetate to remove impurities. The aqueous layer was basified with KOH solution and extracted with DCM. The organic layers were combined and dried over sodium sulfate. The final product amino (4-methoxyphenyl) acetic acid was obtained after evaporation of the solvent under vacuum. The yield was 88%. |
